ATG4B is a cysteine protease that is part of the autophagin protein family. Autophagy is the process by which intracellular proteins and damaged organelles are destroyed. This process is important for cell homeostasis and remodeling during various cellular activities such as differentiation, metamorphosis, non-apoptotic cell death, and aging. ATG4B plays a crucial role in autophagy by cleaving the C-terminal part of proteins MAP1LC3, GABARAPL2, or GABARAP, resulting in the release of form I. Some form I proteins are then transformed into a smaller form called form II. Form II, which has an exposed C-terminal glycine, is known as the phosphatidylethanolamine (PE)-conjugated form and can bind to autophagosomes. Reduced autophagy levels are observed in certain types of malignant tumors, indicating that autophagy may play a role in controlling unregulated cell growth associated with cancer.
