ANGPTL3 and ANGPTL4, secreted mostly by the liver, regulate triglyceride metabolism by inhibiting lipolysis of triglyceride-rich lipoproteins. These proteins play a role in adjusting circulating triglyceride levels during feeding and fasting by affecting Lipoprotein lipase (LPL) activity. Experimental data demonstrates that ANGPTL3 shares similarities with angiopoietins. Deletion mutants of human Angiopoietin 5 indicate that the N-terminal domain (fragment 17-207) rather than the C-terminal fibrinogen-like domain (fragment 207-460) raises plasma triglyceride levels in mice.
