Agouti-related protein is an endogenous antagonist of hypothalamic alpha-melanocortin receptors MC3R and MC4R, demonstrating orexigenic activity. Reduction of AGRP expression through RNA interference has been linked to increased metabolic rate and reduced weight gain. AGRP is primarily produced by neurons in the arcuate nucleus of the hypothalamus, along with the orexigenic Neuropeptide Y (NP-Y). AGRP contains 132 amino acid residues, with its alpha-melanocortin inhibiting activity located in a 34 amino acid cystine knot domain in the C-terminal portion of the protein. Leptin has been shown to suppress both AGRP and NP-Y expression. Central administration of AGRP induces hyperphagia and weight gain in rodents, but may also impact metabolism independently of increased food intake. Peripherally, AGRP mRNA has been found in various tissues, including the adrenal glands, where it has been shown to antagonize glucosteroid production mediated by MC4R. Serum levels of AGRP have been detected in humans and rodents, with elevated levels in obese individuals and an increase following fasting in rats.
