ADAM12 belongs to the A Disintegrin and Metalloprotease protein family, which consists of membrane-anchored proteins closely related to snake venom disintegrins. These proteins play a role in various biological processes involving interactions between cells and their surrounding matrix, such as fertilization, muscle development, and neurogenesis. ADAM12 exists in two forms due to alternative splicing of its transcripts. The shorter variant, known as the S-isoform, is soluble and secreted, while the longer variant, called the L isoform, is membrane-bound. The S-isoform has been found to promote myogenesis, the formation of muscle tissue. Unlike the L isoform, the S isoform lacks the transmembrane and cytoplasmic domains. The expression of ADAM12 S isoform is mainly found in the placenta, embryo, and fetus, although it has also been detected in certain tumor cell lines. ADAM12 is involved in the process of skeletal muscle regeneration, particularly during the initial stages of cell fusion. Additionally, it plays a role in the formation of macrophage-derived giant cells (MGC) and osteoclasts from mononuclear precursor cells, suggesting its involvement in these cellular processes as well.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
