ACE-2 is an enzyme present on cell membranes in various organs like the intestines, arteries, lungs, heart, and kidneys. It serves as an entry receptor for SARS coronaviruses, including SARS-CoV-2. The spike glycoprotein on the coronavirus plays a crucial role in viral infection by binding to host cell receptors and facilitating fusion of viral and cellular membranes. In the S1 domain of the spike protein, two main regions have been identified - the N-terminal domain and C-terminal domain. Both SARS-CoV and MERS-CoV use the C-domain to attach to their receptors. ACE-2 is a transmembrane protein with an extracellular N-terminal domain containing the catalytic site and an intracellular C-terminal tail. It acts as a mono-carboxypeptidase, breaking down Ang I to Ang 1–9 and Ang II to Ang 1–7.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
