Alpha-2 Macroglobulin (A2M) is a liver-produced, large plasma protein composed of four identical subunits bound together by -S-S- bonds. It acts as an inhibitor for various proteases, including thrombin, Kallikrein, plasmin, and multiple other types. A2M contains a 35 amino acid "bait" region that proteinases bind to and cleave, resulting in their attachment to A2M. Macrophage receptors recognize this proteinase-A2M complex and eliminate it from the system. A2M also aids in the removal of MMP-2 and MMP-9 from circulation, using scavenger receptors on phagocytes. The levels of A2M in the body increase in nephrotic syndrome, a condition where the kidneys start leaking smaller blood proteins. As A2M is too large to pass through, its concentration remains elevated. Furthermore, increased production of all proteins contributes to the rise in A2M concentration. Chronic renal failure can potentially lead to the formation of amyloid by alpha-2-macroglobulin. A2M levels are also elevated in cirrhosis, pregnancy, and diabetes.

Storage Instruction

Lyophilized A2M protein although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution and filter sterilization A2M can be stored at 4°C for up to 4 weeks. For long term storage and more diluted solutions it is recommended to add a carrier protein (0.1% HSA or BSA). Please prevent freeze-thaw cycles.

Sample Prep Instruction

It is recommended to reconstitute the lyophilized A2M in sterile 18MΩ-cm H₂O.
