4-1BBL, a transmembrane cytokine belonging to the TNF ligand family, acts as a bidirectional signal transducer for the T lymphocyte receptor TNFRSF9. This interaction plays a role in antigen presentation and cytotoxic T cell generation. Activation of 4-1BBR occurs upon antigenic stimulation, leading to the reactivation of anergic T lymphocytes and promoting T cell proliferation. Optimal CD8 responses in CD8 T cells require 4-1BB Ligand. 4-1BBL is expressed in carcinoma cell lines and plays a role in T cell-tumor cell interaction. Additionally, it is expressed in various immune cells, neurons, and astrocytes. The interaction between 4-1BB and TNFRSF9 strongly regulates immunity, particularly in controlling T cell responses as shown in murine studies.
